Downstream

During the downstream process, the product is recovered from the harvested cell culture fluid and purified from impurities (cells, host cell protein, DNA, viruses, endotoxin, aggregates, and other species) while maintaining an acceptable yield.  

Our purification method employs ÄKTA pure FPLC systems, which are compatible with all kinds of techniques, like affinity chromatography (Protein A), ion-exchange chromatography (IEX), or size-exclusion chromatography (SEC). As a complement of FPLC system, ÄKTA flux tangential flow filtration system coupled with an appropriate membrane can be integrated into different stages of the purification workflows such as for mammalian cell harvest and clarification, diafiltration (buffer exchange), or sample concentration. Also, our research work includes the implementation of PAT tools during protein purification.

Moreover, to overcome the limitations (high cost, batch operation, and long processing time) of the currently applied industrial chromatographic separations we are constantly looking for alternative purification methods, like precipitation or aqueous two-phase extraction.
